A hyperthermophilic, anaerobic, dissimilatory Fe(III)-reducing, facultatively chemolithoautotrophic archaeon (strain SBH6(T)) was isolated from a hydrothermal sample collected from the deepest of the known World Ocean hydrothermal fields, Ashadze field (12 degrees 58' 21 '' N 44 degrees 51' 47 '' W) on the Mid-Atlantic Ridge, at a depth of 4100 m. The strain was enriched using acetate as the electron donor and Fe(III) oxide as the electron acceptor. Cells of strain SBH6(T) were irregular cocci, 0.3-0.5 mu m in diameter. The temperature range for growth was 50-85 degrees C, with an optimum at 81 degrees C. The pH range for growth was 5.0-7.5, with an optimum at pH 6.8. Growth of SBH6(T) was observed at NaCl concentrations ranging from 1 to 6 % (w/v) with an... |

DNJ, an inhibitor of α-glucosidase, is used to suppress the elevation of postprandial hyperglycemia. In this study, we focus on screening an appropriate microorganism for performing fermentation to improve DNJ content in mulberry leaf. Results showed that Ganoderma lucidum was selected from 8 species and shown to be the most effective in improvement of DNJ production from mulberry leaves through fermentation. Based on single factor and three factor influence level tests by following the Plackett-Burman design, the optimum extraction yield was analyzed by response surface methodology (RSM). The extracted DNJ was determined by reverse-phase high performance liquid chromatograph equipped with fluorescence detector (HPLC-FD). The results of RSM showed that the... |

P>Two novel, thermophilic piezophiles, capable of chemolithoautotrophic growth, are successfully cultivated and isolated from a black smoker chimney at the TAG field (Mid Atlantic Ridge: MAR) by using a piezophilic cultivation technique. Both strains (strains 106 and 108) represent dominant cultivated populations of the microbial communities in the chimney surface habitat. Strain 106 represents typically thin, long spiral cells under the piezophilic growth condition but short bent cells under the non-piezophilic condition. It is a strictly chemolithoautotrophic gammaproteobacterium using reduced sulfur compounds as the electron donors, and nitrate and O-2 as the electron acceptors. Based on the 16S rRNA gene sequence, strain 106 would represent a novel... |

Compaction is one of the most destructive factors of soil quality, however the effects on the microbial community and enzyme activity have not been investigated in detail so far. The objective of this study was to evaluate the effects of soil compaction caused by the traffic of agricultural machines on the soil microbial community and its enzyme activity. Six compaction levels were induced by tractors with different weights driving over a Eutrustox soil and the final density was measured. Soil samples were collected after corn from the layers 0-0.10 and 0.10-0.20 m. The compaction effect on all studied properties was evident. Total bacteria counts were reduced significantly (by 22-30 %) and by 38-41 % of nitrifying bacteria in the soil with highest bulk... |

Sulfentrazone is amongst the most widely used herbicides for treating the main crops in the State of São Paulo, Brazil, but few studies are available on the biotransformation of this compound in Brazilian soils. Soil samples of Rhodic Hapludox soil were supplemented with sulfentrazone (0.7 µg active ingredient (a.i.) g-1 soil) and maintained at 27ºC. The soil moisture content was corrected to 30, 70 or 100 % water holding capacity (WHC) and maintained constant until the end of the experimental period. Herbicide-free soil samples were used as controls. Another experiment was carried out using soil samples maintained at a constant moisture content of 70% WHC, supplemented or otherwise with the herbicide, and submitted to different temperatures of 15, 30 and... |

Water used in hydrotherapy units of Nova Iguaçu and Nilópolis, Rio de Janeiro State, Brazil, was microbiologically analyzed. Thirty samples (5ml each) were weekly collected from September 2001 to June 2002 before the beginning and after the end of activities in the units. For analysis, routine techniques were used, which showed the presence of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella spp, Candida spp, Penicillium spp, Epidermophyton spp, Aspergillus fumigatus, Aspergillus niger, Aspergillus spp, Cephalosporium spp, Cladosporium spp, Trichophyton rubrum, and Trichophyton spp. Results indicated a need for improving hygienic conditions, suggesting that water might be a contamination source in the evaluated units. |

Abstract The objective of this work was to evaluate the microbiological attributes of a Plinthossol cropped with sugar cane in straw. The experiment was installed in União (04 ° 51’09”S and 42 ° 53’10 ‘’ W, altitude 52 m), northern region of Piauí state, Brazil. The experimental design was randomized blocks, with four replications, and treatments corresponding to different rates of straw (Mg.ha-1) (0, 4.19, 9.54, 13.04 and 18.38). Microbiological attributes were determined at a depth of 0-10 cm. Data was submitted to analysis of variance, cluster analysis and main components. An increase of straw in the soil influenced the stocks and contents of C, Cmic, Nmic, respiration and microbial and metabolic quotients. Cluster analysis combined the treatments into... |

The author studies the microbial phenomena which occur during the maturation of French anchovies, some from the Basque, the others from the Roussillon coast. There are some differences in relation with the origin of the fishes. On a marine agar, the microflora of the Mediterranean ones is almost constant or in a light diminution while that of the Atlantic ones is climbing during 9 weeks before a decline. On an extremely salted agar and in the both origins the archaebacteria are increasing in number during 5 to 6 weeks and diminishing before. The complete ripening is obtained within 16-17 weeks. The limits of the significance of the microbiological analysis are discussed. |
