Genetic variability of Brazilian phytoplasma and spiroplasma isolated from maize plants PAB
Gomes,Eliane Aparecida; Jardim,Sílvia Neto; Guimarães,Claudia Teixeira; Souza,Isabel Regina Prazeres de; Oliveira,Elizabeth de.
The objective of this work was to characterize the genetic variability of phytoplasma and Spiroplasma kunkelii isolated from maize plants showing symptoms of stunt collected from different Brazilian geographic regions. A DNA fragment of 500 base pairs (bp) was amplified from the spiralin gene in S. kunkelii and one fragment of 1,200 bp was generated from 16S rDNA gene in phytoplasma. The partial sequences of the spiralin gene showed similarity of 98% among the isolates of S. kunkelii analyzed. These sequences were compared with the sequence of the spiralin gene from other Spiroplasma species deposited in the GenBank, resulting in a similarity varying from 76.9% to 88.1%. The 16S rDNA sequence from the phytoplasma were completely similar within the...

Improved 1-Deoxynojirimycin (DNJ) production in mulberry leaves fermented by microorganism BJM
Jiang,Yun-Gang; Wang,Chu-Yan; Jin,Chao; Jia,Jun-Qiang; Guo,Xijie; Zhang,Guo-Zheng; Gui,Zhong-Zheng.
DNJ, an inhibitor of α-glucosidase, is used to suppress the elevation of postprandial hyperglycemia. In this study, we focus on screening an appropriate microorganism for performing fermentation to improve DNJ content in mulberry leaf. Results showed that Ganoderma lucidum was selected from 8 species and shown to be the most effective in improvement of DNJ production from mulberry leaves through fermentation. Based on single factor and three factor influence level tests by following the Plackett-Burman design, the optimum extraction yield was analyzed by response surface methodology (RSM). The extracted DNJ was determined by reverse-phase high performance liquid chromatograph equipped with fluorescence detector (HPLC-FD). The results of RSM showed that the...

Interactions of deep-sea vent invertebrates with their environment: The case of Rimicaris exoculata ArchiMer
Schmidt, Caroline; Le Bris, Nadine; Gaill, F.
The vent shrimp Rimicaris exoculata thrives around many hydrothermal vent sites along the Mid-Atlantic Ridge (MAR), where it aggregates into dense swarms. In contrast to hydrothermal vent fields at the East Pacific Rise (EPR), where the biomass is dominated by tubeworms, clams, and mussels, this shrimp is one of the major animal species at MAR vents. These animals are found in the dynamic mixing interface between cold oxygenated seawater and hot, reduced hydrothermal vent fluid. The adaptation of this shrimp to the hostile deep-sea hydrothermal environment and its survival strategy has been investigated since their discovery at the TAG site in the late 1980s. Rimicaris exoculata is now known to colonize black smoker complexes along the MAR in the...

Isolation and physiological characterization of two novel, piezophilic, thermophilic chemolithoautotrophs from a deep-sea hydrothermal vent chimney ArchiMer
Takai, Ken; Miyazaki, Masayuki; Hirayama, Hisako; Nakagawa, Satoshi; Querellou, Joel; Godfroy, Anne.
P>Two novel, thermophilic piezophiles, capable of chemolithoautotrophic growth, are successfully cultivated and isolated from a black smoker chimney at the TAG field (Mid Atlantic Ridge: MAR) by using a piezophilic cultivation technique. Both strains (strains 106 and 108) represent dominant cultivated populations of the microbial communities in the chimney surface habitat. Strain 106 represents typically thin, long spiral cells under the piezophilic growth condition but short bent cells under the non-piezophilic condition. It is a strictly chemolithoautotrophic gammaproteobacterium using reduced sulfur compounds as the electron donors, and nitrate and O-2 as the electron acceptors. Based on the 16S rRNA gene sequence, strain 106 would represent a novel...

Microbial alterations of the soil influenced by induced compaction Rev. Bras. Ciênc. Solo
Pupin,Breno; Freddi,Onã da Silva; Nahas,Ely.
Compaction is one of the most destructive factors of soil quality, however the effects on the microbial community and enzyme activity have not been investigated in detail so far. The objective of this study was to evaluate the effects of soil compaction caused by the traffic of agricultural machines on the soil microbial community and its enzyme activity. Six compaction levels were induced by tractors with different weights driving over a Eutrustox soil and the final density was measured. Soil samples were collected after corn from the layers 0-0.10 and 0.10-0.20 m. The compaction effect on all studied properties was evident. Total bacteria counts were reduced significantly (by 22-30 %) and by 38-41 % of nitrifying bacteria in the soil with highest bulk...

Martinez,Camila O.; Silva,Celia Maria M. S.; Fay,Elisabeth F.; Abakerli,Rosangela B.; Maia,Aline H. N.; Durrant,Lucia R..
Sulfentrazone is amongst the most widely used herbicides for treating the main crops in the State of São Paulo, Brazil, but few studies are available on the biotransformation of this compound in Brazilian soils. Soil samples of Rhodic Hapludox soil were supplemented with sulfentrazone (0.7 µg active ingredient (a.i.) g-1 soil) and maintained at 27ºC. The soil moisture content was corrected to 30, 70 or 100 % water holding capacity (WHC) and maintained constant until the end of the experimental period. Herbicide-free soil samples were used as controls. Another experiment was carried out using soil samples maintained at a constant moisture content of 70% WHC, supplemented or otherwise with the herbicide, and submitted to different temperatures of 15, 30 and...

Microflore et anchoitage. ArchiMer
Campello, Francois.
The author studies the microbial phenomena which occur during the maturation of French anchovies, some from the Basque, the others from the Roussillon coast. There are some differences in relation with the origin of the fishes. On a marine agar, the microflora of the Mediterranean ones is almost constant or in a light diminution while that of the Atlantic ones is climbing during 9 weeks before a decline. On an extremely salted agar and in the both origins the archaebacteria are increasing in number during 5 to 6 weeks and diminishing before. The complete ripening is obtained within 16-17 weeks. The limits of the significance of the microbiological analysis are discussed.

Microflores sulfatoreductrices en milieu subantarctique. Relations avec quelques parametres physicochimiques et biologiques. (Microflores totales et heterotrophes) ArchiMer
Delille, D; Bouvy, M.
The archipelago of Kerguelen with its complex fjord distribution is particularly interesting for the study of anoxic systems. Between 1968 and 1972, several stations were investigated fortnightly. One of these was further sampled weekly throughout 1982. Each sample series of sediment included the superficial aerobic and subsuperficial anaerobic levels. Each level was examined for direct epifluorescent counts, heterotrophic and sulfate-reducing bacterial numbers by M.P.N. methods, organic carbon and sulfide measurements. Sulfate-reducing bacteria seemed less related to total biomass than the heterotrophic microflora, the latter being dependent on organic supplies. The hydrogen sulfide production was directly related to the presence of sulfate reducers;...

Microrganismos e disponibilidade de fósforo (P) nos solos: uma análise crítica. Infoteca-e
MENDES, I. de C.; REIS JUNIOR, F. B. dos.
ABSTRACT: Cerrado soils are extremely old, highly weathered and depleted in essential nutrients, especially P. For this reason, the addition of P fertilizers is mandatory in order to obtain economical yields of annual crops. Soil microorganisms play a key role in the P cycle. The P immobilized in the soil microbial biomass acts as reservoir of this element. Soil microorganisms also produce enzymes that act in the P mineralization, participate in the solubilization of inorganic phosphorus and are able to form symbiosis with plants, denominated mycorrizha, enhancing the uptake of this element. In this review we discuss the results of several studies evaluating the use of soil microorganisms to increase P availability in soils, some of the difficulties...

Models of relations between bacteria and plankton, and regeneration processes in marine planktonic ecosystem. ArchiMer
Martin, Y; Lelong, P; Cauchi, B.
In order to study bacteria-plankton relationships, experimental and mathematical models were used to stimulate biological processes implicated in plankton ecosystems and to analyze community relationships. The model components are phytoplankton, zooplankton, autotrophic and heterotrophic bacteria, ciliates, nutrients, organic matter and phytoplankton antibiotics. This study shows the importance of some factors affecting bacteria communities dynamics and the bacterial activity in mineralization process. Bacterial populations seem to be controlled essentially by the concentration and chemical composition of organic substrates. Antibiosis and predation show a lesser importance on changes in bacteria communities.
